Mahoney started onstage as a child, where she was bitten by the acting bug. She went on to receive her B.A. in Theatre Arts from Greensboro College, and her M.F.A. in Acting from the University of Connecticut. Her undergraduate work focused on Western acting techniques, including Stanislavski & Meisner. Her graduate work was primarily focused on The Suzuki Method of Actor Training, an extreme physical and vocal training technique created by Japanese Theatre Master, Tadashi Suzuki.

Mahoney has been seen Off-Broadway and in regional theatres around the country including, Long Wharf Theatre, The Berkshire Theatre Festival, and Southern Rep. Known as a versatile actor, Ann is best remembered for her role as "Olivia" on three seasons of The Walking Dead. Selected TV credits include the iconic Gladys Presley in Sun Records, and Guest Starring roles on The Sundance Channel's Rectify, the CW's Hart of Dixie, and The Good Cop for Netflix. Her Film appearances include Bad Moms, Logan Lucky, 99 Homes, and Same Kind of Different as Me, among others. The book draws on Mahoney's 25 years experience both onstage and on camera, as well as 10 years of teaching Acting for Camera on the university level. Mahoney specializes in "telling it like it is," and giving theatre actors the skills and inspiration to become compelling on-camera actors.
